Determination of Test Results. 6. Incubate this test for 24 hours. Retrieve the incubated culture. Look for the presence or absence of a zone of inhibition around the disk. Interpret these results. 7. Select “Record Results” option when clicking on the plate. Report the results and dispose of the medium.
this test Nitrate • Red/pink color after reagents/no color after zinc + • No color change/red after zinc – • Nitrate Reagents A & B • Reduction of nitrate to nitrite to be used as a final electron acceptor/Nitrate reductase •NO 3-reductase results: E. coli +, S. epidermidis +, soil -, P. fluorescens-, M.luteus-
Published: Sulfate-reducing bacteria (SRBs) gain their energy by coupling the oxidation of organic substrate to the reduction of sulfate to sulfide. Several SRBs are able to use alternative terminal electron acceptors to sulfate such as nitrate. Nitrate-reducing SRBs have been isolated from a diverse range of environments.
E.coli - Positive w/ A&B solution (red) end product of NO2- (positive Nitrate reduction test) P.aeruginosa- negative w/ A&B solution and positive for Nitrate Reductions of either NH3 or N2 (no color change/or could say yellow from zinc)
E. coli. respiratory nitrate reductase. E. coli NarGH activates the first time it is reduced, through a sequential two-step process. The inactive species are not related to Mo (V) high-pH or low-pH signatures, nor to FS0 or FS1 signatures. The high-pH and low-pH Mo (V) species are not in an acid/base equilibrium.
I. Oxidative-fermentative test using OF media with glucose A. Inoculation of media Two tubes of oxidative-fermentative medium are inoculated by stabbing “half way to the bottom” (8) or ¼-inch from the bottom (6) with the test organism. Overlay one of the two tubes with 1 cm of mineral oil (8) (Fig. 1, 2, 3).
